How to create MRM methods and analysis? Kynda  2017-10-31 15:54

Hi,
I am new to proteomics.

I want to perform MRM for organelle specific proteins and some pathway specific proteins. There are around 80 interested target proteins.
For some of the proteins (Gene IDs /AGI) I have qTOF libraries (around 20). So for others I have to create theoretical digestions as I got to know.

Could you please please help me to find guidelines/tutorials for this.

What are the steps I should follow to create the methods for MRM and how can screen my samples after that.

Before screen the sample I got to know I have to create pool and screen the targeted proteins?

Hi Kynda,

I recommend checking out the resources on the Skyline web site. We support the Panorama module, which accepts data from Skyline to help analyze and share it, but that's downstream of the initial MRM setup.

https://skyline.ms/project/home/software/Skyline/begin.view

They have good tutorials for how to use Skyline for MRM.

Thanks,
Josh
